Trypsin Laboratory

  1. Purposes:
    1. To investigate optimal conditions for trypsin activity (pH and temperature).
    2. To apply tertiary structure theory to understand behavior of trypsin.

  2. Introduction:
    • Suggestions: Give some background on trypsin. (Where produced, structure and what it's made of, any extra information to help a reader understand the discussion later.)
    • Why was trypsin selected? (Ex: Can be stored in powder form for years.)

  3. Hypothesis:
    One sentence (or two) = prediction

  4. Data (Results):
    Make an observation and interpretation (the why's for observation) chart.
    • Your observations should be phrases, clauses and/or brief sentences.
    • Make this chart so some of it can be used, after elaborating, in the paragraphs of the discussion.

  5. Methods and Materials:
    • Use past tense.
    • Explain how the lab was set up.
    • What was the control?
    • What were the ariables in the procedure?
    • Don't copy out the procedure, just make reference to it.

  6. Procedure:
    1. Place a cubic cm of boiled egg white in each of the six test tubes.
    2. Add four cubic cm of trypsin solution and buffer solution to each of the test tubes.
      1. Add water to tube #1.
      2. Add buffer solution pH2 to a sample of trypsin. Add this mixture to tube #2.
    3. Place tubes 1,2,3 and 4 in the incubator (37°C).
    4. Obtain a fifth test tube and place pH8 buffer and trypsin and egg white in it. Place it in the fridge.
    5. Add only pH buffer 8 to Test Tube #6. Why is this tube a second control?
    6. Try an extra procedure. (Ask first)! ex: Boil trypsin first.

  7. Discussion:
    1. Explain how the theory of tertiary structure of proteins relates to the results.
    2. Explain how the lock and key model of enzyme action relates to the results.
    3. Explain how various conditions relate to the results (HABS)
    4. Why was buffer added?
    Suggestions: Significance and Implications Ex: Suppose somebody had a serious pancreatic disease. What would the results and symptoms be (Re: digestion) and why?

  8. Conclusion:
    Two or three general overall statements about what you got out of the lab regarding the results.
